- Stauprimide is used in life sciences, particularly for biological activity research. It promotes embryonic stem cell (ESC) differentiation by binding to MYC transcription factor NME2 and blocking its nuclear localization, leading to MYC transcription downregulation. In vitro, it suppresses MYC transcription in various cell lines with EC50 30 nM-8 μM, decreases MYC levels, inhibits cell proliferation (IC50 780 nM in RXF 393 cells), and reduces nuclear NME2. In vivo, oral administration at 50 mg/kg once daily for 30-55 days blocks tumor growth in xenograft mouse models with RXF 393 or CAKI-1 cells, reducing MYC protein levels and inhibiting MYC transcription in tumors.
