PIM447 dihydrochloride is a selective pan-PIM kinase inhibitor for treating multiple myeloma (MM). It inhibits MM cell viability (IC50 0.2-3.3 µM in sensitive lines), induces apoptosis via caspase cleavage and PARP cleavage, arrests cell cycle in G0/G1 phase, and controls tumor progression with bone protective effects in NSG mice (oral gavage, 100 mg/kg, 5x/week).
