- Deramciclane 是一种新型抗焦虑药物,对 5-HT2A 和 5-HT2C 受体具有高亲和力,作为两种受体亚型的拮抗剂,对 5-HT2C 受体具有反向激动剂特性,无直接刺激激动剂。在体外研究中,其通过受体磷酸肌醇水解测定和受体放射自显影进一步表征与 5-HT2C 受体的相互作用,对 5-HT2C 受体介导的 5-HT 刺激磷酸肌醇水解的拮抗 IC50 值为 168 nM,在脉络丛的生理系统中可降低基础磷酸肌醇水解高达 33%(EC50 = 93 nM),表明具有反向激动剂特性。在体内研究中,Deramciclane 3 和 10 mg/kg 对多巴胺水平无显著影响,30 mg/kg 时在 40-100 min 和 160-240 min 显著增加多巴胺水平(P<0.05),是一种假定的抗血清素能化合物,可减少 5-HT 诱导的磷酸肌醇水解及多种血清素能激动剂引起的作用,其受体结合谱与利坦色林相似,在几种动物试验中显示出抗焦虑样活性。
