Jervine (11-Ketocyclopamine)是Hedgehog信号通路的抑制剂(IC50=500-700 nM),通过与smoothened相互作用抑制shh信号通路。

- 体外研究
相较于对照组,暴露于25 μg/ml jervine的软骨细胞在细胞数目上有明显的减少,而在细胞形态或阿尔新蓝染色的强度没有变化。
- 体内研究
暴露于jervine的胚胎会产生颅面骨畸形,与Shh突变胚胎表型类似。这些胚胎的下颌在大小上显著减小,通常会造成门齿的缺失。对受孕小鼠处理以jervine会导致其胚胎发生颌骨缺损,其表型与Prx1。
