Atrasentan (ABT-627) is a pharmaceutical compound used as an endothelin receptor (ETA) antagonist with IC50 0.055 nM. It inhibits LNCaP and C4-2b prostate cancer cell growth, synergizes with Taxotere, downregulates NF-κB, induces CYP3A4 and drug transporters, and inhibits P-gp (IC50 15.1 μM) and BCRP (IC50 59.8 μM). In vivo, it inhibits big endothelin-1 pressor response and C4-2b tumor growth in bone environment.
