合成条件:Stage #1: With sodium hydride In N,N-dimethyl-formamide; mineral oil at 20℃; for 0.50 h; Stage #2: at 20℃;
实验步骤:实施例13A 2-(4-溴苯基)-2-甲基丙酸乙酯将2.0g(8.2mmol)乙酸(4-溴苯基)乙酸酯溶于50ml DMF和0.7g(18mmol)氢化钠矿物油(在室温下加入60%)。将混合物在室温下搅拌30分钟,然后加入2.9g(20.6mmol)碘甲烷。然后将混合物在室温下搅拌过夜。将混合物用乙酸乙酯稀释,先用水萃取,然后用饱和氯化钠溶液萃取。分离出有机相,用硫酸镁干燥并过滤,浓缩滤液。通过制备型HPLC [ReprosilC18,10μm,250mm×40mm(30%甲醇/ 70%水至100%甲醇),在25分钟的运行时间]纯化残余物。 HPLC控制后,合并含有产物的级分并浓缩。得到1.75g(理论值的78%)油。 GC-MS [方法5]:Rt = 5.10分钟; MS(ESIpos):m / z = 270/272(M +)1H-NMR(400MHz,DMSO-d6):δ[ppm] = 1.11(t,3H),1.48(s,6H),4.06(q, 2H),7.24-7.29(m,2H),7.50-7.54(m,2H)
