实验步骤:步骤B:2,4,7-三氯喹唑啉的制备。 将7-氯-1H-喹唑啉-2,4-二酮(2.0g,10mmol)的混合物悬浮在ACN(50mL)中,然后加入POCl 3(5.0mL,55mmol)。 然后加入DIEA(5.0mL,28mmol)。 将所得混合物加热至回流36小时,然后冷却至室温并浓缩。 用冰和碳酸氢钠小心处理残余物。 过滤收集所得固体并干燥。 色谱纯化(EtOAc /己烷0:100至10:90)得到标题化合物(2.1g,89%)。 MS和NMR数据与先前描述的那些一致:Bioorganic and Medicinal Chemistry,2003,11,2439-2444。 1H NMR(400MHz,DMSO-d6):8.32(d,J = 8.7Hz,1H),8.20(d,J = 1.9Hz,1H),7.93(dd,J = 9.0,2.1Hz,1H)。
