实验步骤:5-溴I-(三异丙基硅烷基)-1H-吡咯并[2,3-b]吡啶:; 在室温下将氢化钠(0.63g,25.15mol)分小份加入到搅拌的5-溴-7-氮杂吲哚(3.3g,16.75mmol)的THF(50mL)溶液中,并在室温下搅拌所得悬浮液。 温度15分钟。 加入三异丙基甲硅烷基氯(5.3mL,25.15mmol),将混合物在80℃加热3小时。 蒸发溶剂,将残余物溶于水(50mL)中。 将水层用EtOAc(3×50mL)萃取,干燥(Na 2 SO 4)并减压浓缩。 通过Biotage HPFC系统(10%EtOAc /己烷)纯化粗产物,得到所需的5-溴-1-(三异丙基甲硅烷基)-1H-吡咯并[2,3-b]吡啶,为白色固体(5.2g,88%)。。; 质谱355(M + 1)。

实验步骤:第4步; 向搅拌的5-溴-1H-吡咯并[2,3-b]吡啶(1.5克,7.5毫摩尔)在15毫升二恶烷和5毫升DMF中的溶液中加入氢化钠(345毫克,9毫摩尔)。 分批,0.5小时后,TIPSCI(2.5毫升,12.3毫摩尔); 在室温下搅拌反应物。 过夜,然后用水稀释并用二氯甲烷萃取; 用二氯甲烷通过硅胶色谱法快速纯化,得到2.6g 5-溴-1-三异丙基硅烷基-1H-吡咯并[2,3-b]吡啶(4)(产率97%)。 该产物无需进一步纯化即可用于下列反应。
