合成条件:With sodium hydroxide In water at 0 - 20℃;
实验步骤:将L-色氨酸(10.24g,50mmol)溶解在1M NaOH(50mL)中并在0℃下搅拌。 氯甲酸苄酯(7.15mL,50.1mmol)和1M NaOH(50mL),然后以逐滴方式同时加入。 将混合物在室温下搅拌1小时。 将溶液用6M HCl酸化至pH 1,然后用EtOAc(3×200mL)萃取产物。 合并有机层,用Na 2 SO 4干燥并蒸发。 得到黄色固体产物,直接用于下一步骤(16.72g,99%)。 分析数据:Rf 0.43(95:4:1 CH 2 Cl 2 / MeOH / AcOH); Mp:132-134ºC; 1H NMR(DMSO-d6):δ7.57-7.53(m,2H),7.35-7.22(m,6H),7.16-7.15(m,1H),7.09-7.04(m,1H)7.00-6.95(m, 1H),5.02-4.92(m,2H),4.28-4.21(m,1H),3.22-3.16(m,1H),3.04-2.96(m,1H); 13C NMR(DMSO-d6):δ174.2,156.5,137.4,136.6,128.8,128.2,128.0,127.6,124.2,121.4,118.8,118.6,111.9,110.5,65.8,55.5,27.4; LRMS(ESI):C 19 H 19 N 2 O 4 [M + H] +计算值339.13,实测值339.35。
