合成条件:Stage #1: With sulfuric acid; sodium nitrite In water at 0 - 5℃; for 1 h; Stage #2: at 100℃; for 0.50 h;
实验步骤:将亚硝酸钠(90mL的0.5M水溶液)加入冰冷搅拌的5-氨基-2-氯苯甲酸(16)(5.01g,29.6mmol)和硫酸(6.00mL,0.167mol)的悬浮液中。在水(460毫升)中,在加入过程中保持温度在0和5℃之间。搅拌1小时后,将反应混合物用水(900mL)稀释,然后在回流下再搅拌30分钟。然后将反应混合物冷却至室温,并萃取到乙酸乙酯(3×500mL)中。将合并的有机相干燥(MgSO 4),过滤并减压浓缩,得到17,为浅棕色固体(4.97g,98%收率)。熔点熔点165-168℃; IR(ZnSe):3311,2920,2598,1676,1599,1573,1474,1447,1228,826cm -1; 1H NMR(300MHz,MeOD):δ7.26(1H,d,J = 8.7Hz),7.23(1H,d,J = 3.0Hz),6.89(1H,dd,J = 3.0和8.7Hz)ppm 2xOH信号没有观察到; 13C NMR(75MHz,MeOD):δ169.1,157.4,132.7,132.8,123.8,120.7,116.2ppm; LRMS(-ESI):171([M-H] - ,100%),127([M-CO2H] - ,36%)。
