合成条件:Stage #1: With sodium hydride In tetrahydrofuran; mineral oil at 0℃; for 0.50 h; Inert atmosphere Stage #2: at 0 - 20℃; for 4.50 h; Inert atmosphere
实验步骤:在0℃下,向Boc-L-Leu(5.0g,21.6mmol)的THF(73mL)溶液中加入NaH(60%在矿物油中的分散体,10.0g,0.13mol)。 在0℃下搅拌30分钟后,依次加入MeI(4.1mL,66.0mmol)和DMF(3.7mL)。 将反应在0℃下搅拌30分钟并在室温下再搅拌4小时,直至其用H 2 O(50mL)淬灭。 将混合物用EtOAc(2×100mL)萃取。 然后将水溶液用10%KHSO 4水溶液酸化至pH 5并用乙酸乙酯萃取。 将合并的EtOAc萃取液用5%Na 2 S 2 O 4溶液和盐水洗涤,用Na 2 SO 4干燥,过滤并蒸发,得到13(5.2g,98%收率),为无色油状物。
