合成条件:Stage #1: With potassium permanganate In water at 100℃; for 53 h; Stage #2: With hydrogenchloride In water
实验步骤:合成6-溴-5-氟吡啶甲酸向2-溴-3-氟-6-甲基吡啶(1.0当量)的H 2 O(30mL)溶液中加入高锰酸钾(1.0当量)。 将溶液在100℃下加热5小时,此时加入更多的高锰酸钾(1.0当量)。 再加热48小时后,将物质通过硅藻土(4cm×2英寸)过滤并用H 2 O(150mL)冲洗。 将合并的水溶液用1N HCl酸化至pH = 4,用乙酸乙酯(200mL)萃取,用NaCl(饱和)洗涤,经MgSO 4干燥,过滤并浓缩,得到6-溴-5-氟吡啶甲酸(17%)。 为白色固体。 LCMS(m / z):221.9(MH +); LC Rt = 2.05分钟。
