合成条件:With N-iodo-succinimide In N,N-dimethyl-formamide at 20 - 65℃;
实验步骤:在室温下将1.5当量的N-碘代琥珀酰亚胺加入到25g二甲基甲酰胺中的5g(32.7mmol)3-氨基吡嗪-2-羧酸甲酯溶液中。 将反应介质在65℃加热1小时,与0.5当量的N-碘代琥珀酰亚胺一起加入,并在65℃下保持24小时。 回到室温后,蒸发溶剂,然后用二氯甲烷萃取产物数次。 合并有机相,用10%亚硫酸氢钠溶液洗涤,用硫酸镁干燥并浓缩,得到8g(88%)黄色固体形式的3-氨基-6-碘吡嗪-2-羧酸甲酯.LCMS(EI) m / z):( M + 1)2801H NMR:6H ppm(400MHz,DMSO):8.50(1H,s,CHarom),7.50(2H,bs,NH 2),3.20(3H,s,CH 3)。
