实验步骤:将DMF(6.2mL,80.65mmol)和磷酰氯(8.1mL,88.61mmol)混合,然后加入2-甲基间苯二酚(5.0g,0.806mmol)乙酸乙酯(100mL)。将该溶液在室温下搅拌2小时。 乙酸乙酯,与有机相合并,用无水硫酸钠干燥,过滤,减压浓缩,产物在硅胶柱上色谱分离(4.9g,79.9%);

实验步骤:A.制备2,4-二羟基-3-甲基苯甲醛将磷酰氯(80mL,0.86mol)加入到搅拌的N-甲基甲酰苯胺(102mL,0.82mol)的乙醚(250mL)混合物中。将混合物在室温下搅拌1小时,然后在冰中冷却。加入2-甲基间苯二酚(Aldrich,100g,0.81mol),将混合物温热至室温,同时搅拌过夜。过滤收集沉淀的中间产物,用乙醚(3x)冲洗。通过溶解在丙酮(250mL)和水(250mL)的混合物中水解中间体并搅拌30分钟。加入水(2L),使混合物沸腾,然后冷却并沉积结晶产物。将其从水(4L)中第二次重结晶,得到纯产物(70g,57%)。熔点150℃(Lit.152-3℃[W.Baker等人,J.Chem.Soc。,2834-5(1949)。]。NMR(DMSO-d6):w 1.973(s,3H) ),6.551(d,J = 8.5hz,1H),7.428(d,J-8.5hz,1H),9.703(s,1H),10.745(s,1H)和11.592(s,1H)。
