Xevinapant (AT-406) hydrochloride is an effective, orally bioavailable Smac mimetic and IAPs inhibitor antagonist. It binds to XIAP, cIAP1, and cIAP2 proteins with Ki values of 66.4, 1.9, and 5.1 nM respectively. It effectively antagonizes XIAP BIR3 protein in cell-free functional assays, induces rapid degradation of cell cIAP1 protein, inhibits cancer cell growth in various human cancer cell lines, and is highly effective in inducing apoptosis in xenograft tumor cells.
