路线1:以(4R-顺式)-6-(羟甲基)-2,2-二甲基-1,3-二恶烷-4-乙酸,1,1-二甲基乙基酯为原料合成
- 步骤:将(4R-顺式)-6-(羟甲基)-2,2-二甲基-1,3-二恶烷-4-乙酸,1,1-二甲基乙基酯(10.0g,0.0385mol)溶解在二甲基亚砜(15.0g,0.192mol)和二氯甲烷(100ml)中,冷却至0至-5℃并搅拌15分钟;加入二异丙基乙胺(17.42g,0.1347mol),在0至-5℃下继续搅拌15分钟;将吡啶-三氧化硫复合物(12.25g,0.077mol)、吡啶(6.09g,0.077mol)和二甲基亚砜(15.0g,0.192mol)室温搅拌10分钟得到悬浮液,在0至-5℃下加入上述醇溶液中,继续搅拌1小时;反应完成后加入水(50.0ml)搅拌10分钟,分层后水层用二氯甲烷(2×20ml)萃取,合并的二氯甲烷层用水(3×100.0ml)洗涤,经硫酸钠干燥后减压蒸馏(40至45.0℃)得到产物。
- 条件:Stage #1:使用二甲基亚砜、N-乙基-N,N-二异丙基胺,在二氯甲烷中于-5至0℃反应0.50小时;Stage #2:使用吡啶、三氧化硫吡啶复合物、二甲基亚砜,在二氯甲烷中于-5至20℃反应1.17小时。
