实验步骤:将氢氧化钠(40mL,6.25M溶液)加入搅拌的4,6-二氯烟酸乙酯(22)(25.95g,118mmol)在4:1:1 THF:MeOH:水(600mL)中的溶液中。。 30分钟后,将反应混合物用浓HCl酸化至pH 2,用1:1 EtOAc:Et 2 O稀释,并用水和盐水洗涤。 将有机层干燥(Na 2 SO 4)并浓缩。 将得到的灰白色固体从甲苯中浓缩两次,得到所需产物(23),为白色固体(21.73g,96%)。

实验步骤:将4,6-二氯 - 烟酸甲酯(27.3g,133mmol)溶解在THF(150mL)中。 逐滴加入溶解在H 2 O(25mL)中的LiOH(3.53g,147mmol)并将反应搅拌18小时。 真空除去挥发物。 将得到的残余物用H 2 O(100mL)稀释,并用6N HCl酸化,同时在0℃下旋转。使溶液的pH为2.0,形成黄色沉淀。 将混合物在0℃下静置1小时,然后过滤,得到4,6-二氯 - 烟酸,为黄色固体(20.6,81%)。
