实验步骤:将化合物28(15.0g,0.0738mol)和二碳酸二叔丁酯(17.7g,0.0812mol)溶解在EtOAc(400ml)中。 加入氢氧化钯催化剂(4g)并将混合物在Paar振荡器上在40psi氢气压下振荡24小时。 将所得混合物通过硅藻土过滤,并用EtOAc洗涤硅藻土。 浓缩滤液,得到15.73g(0.0738mol,100%)产物27B,为无色油状物。 MS(M-tBOC的ES + 1):m / e 114

合成条件:Stage #1: With lithium diisopropyl amide In tetrahydrofuran at -78℃; for 0.50 h; Inert atmosphere Stage #2: at -78 - 20℃; for 3 h;
实验步骤:在-78°下,向3-甲基-4-氧代哌啶-1-羧酸叔丁酯(1.0g,5.00mmol)的无水THF(10mL)溶液中滴加LDA(3mL,6.0mmol)。 C在氮气氛下。 30分钟后,在相同温度下将Mel(852mg,6.0mmol)加入上述溶液中,然后将混合物在室温下再搅拌3小时。 将混合物用饱和NH 4 Cl溶液淬灭,并在EtOAc(40mL)和水(20mL)之间分配。 用盐水(10mL)洗涤有机层,经硫酸钠干燥,浓缩,得到橙色油状物。 通过硅胶柱色谱法(石油醚/乙酸乙酯= 20:1)纯化残余物,得到化合物,为无色油状物(500mg,45%)。 1 H NMR(400MHz,CDCl 3)δ4.11(m,1H),3.71(m,1H),3.23(m,1H),2.81(br s,1H),2.47(m,3H),1.47(s, 9H),1.03(d,/ = 6.8Hz,3H)。
