合成条件:With sodium sulfate In tetrahydrofuran at 55 - 60℃; Inert atmosphere
实验步骤:在室温下将2-氨基-4-氯酚(50g,0.349摩尔),原甲酸三甲酯(111g,1.048摩尔),硫酸钠(9.93g,0.069摩尔)和THF(500mL)装入圆底烧瓶中 (RT)在惰性气氛下。 将反应物料加热至55℃至60℃。通过TLC监测反应进程。 反应完成后,将反应混合物冷却至室温并用水(50ml)淬灭。 将反应物料冷却至室温,用水(250ml)稀释,并用乙酸乙酯(2×250ml)萃取。 将合并的有机萃取液用硫酸钠干燥,过滤并减压浓缩。 所得粗物质通过柱色谱法纯化,使用硅胶(60-120目),用20%EtOAc-己烷洗脱,得到5-氯-1,3-苯并恶唑(式XI,50g,收率95%,HPLC纯度99.46%)。 )为黄色固体。
