- 合成条件:Stage #1: 以叔丁醇钾为试剂,四氢呋喃为溶剂,在20℃、惰性气氛下反应8小时;Stage #2: 加入硫酸,在四氢呋喃中20℃反应18小时
- 实验步骤:中间体1:2-氧代环戊烷甲腈在室温和氮气氛下,将叔丁醇钾(41.51g,370mmol)加入搅拌的己二腈(426mL,370mmol)和THF(600mL)溶液中。搅拌8小时后,向反应中加入H₂F₄(19.72mL,370mmol)(1M溶液),并将得到的混合物在室温下搅拌18小时。加入NH₄Cl(600mL)并将所得混合物用EtOAc(2×500mL)萃取。将合并的有机层经Na₂SO₄干燥并真空除去溶剂,得到2-氧代环戊烷甲腈(29.31g,269mmol,73%收率),为黄色油状物。¹H NMR(CDCl₃,400MHz)O/ppm:3.20(1H,dd,J=8.4Hz,10.2Hz),2.15-2.60(5H,bm),1.92(1H,m)。MS方法2:RT:0.76ml mlz 110.1 [M + H]
