路线1:以(2R,4R)-1-甲基4-羟基吡咯烷-1,2-二甲酸叔丁酯为原料
- 条件:重铬酸吡啶鎓(9.97g,26.5mmol)、二氯甲烷(60mL),室温
- 步骤:将重铬酸吡啶鎓二氯甲烷溶液加入原料中,室温搅拌过夜;加入硅藻土搅拌40分钟后过滤,滤饼用二氯甲烷冲洗;浓缩滤液,加入甲苯再次浓缩得粗产物;经快速硅胶色谱法纯化
- 产率:84%
- 产物:白色固体
- LCMS:m/z 144(M-Boc)-ve
- ¹H NMR:(400MHz, CDCl3) δppm4.44-4.89(1H,m),3.84-3.96(2H,m),3.77(3H,s),2.81-3.10(1H,m),2.59(1H,dd,J=18.93,1.76Hz),1.52(9H,s)

- 条件:Dess-Martin periodane(20.75g,48.9mmol)、DCM(408ml),室温
- 步骤:将原料悬浮于DCM中,一次性加入Dess-Martin periodane,室温搅拌过夜;用水和DCM稀释后过滤,分离有机层并用水洗涤两次,干燥(MgSO4)后过滤真空浓缩得粗产物;经硅胶色谱法(0-20%EtOAc/庚烷)纯化
- 产物:9.92g,静置后固化
